By default, After Effects will display the first frame of any composition as the poster (thumbnail) image within the Project panel. To change this drag the playhead on the timeline to a particular frame. Choose "Set Poster Time" from the Composition menu to make that frame the poster image.
If a composition contains transparency, this can be shown in the thumbnail. From the Project panel menu select "Thumbnail Transparency Grid" and the checkerboard pattern will be shown wherever there is transparency.
