DNA foot print assay is a technique used to find, region of DNA where a particular protein binds. The protein can be a transcription factor, enhancer or a repressor protein. For this assay the DNA segment is amplified by PCR and then labeled. The test protein is then added to the DNA fragments to be analyzed. If the protein interacts with the DNA then DNAse I enzyme will fail to digest the region where the protein was interacting. This results in foot print on the gel.
