Digital PCR (dPCR) can provide many advantages when implemented into your genetic workflows. However, choosing the right dPCR system is important to get accuracy and precision in your results.
Digital PCR can be done by limiting dilution or the Poisson method. The Poisson method, which required samples be partitioned into thousands of subreactions, is the most common for dPCR. Sample partitioning can be accomplished by separation into microwells or chambers, or by droplet formation using liquid emulsion.
Separation into microwells and chambers sounds easy but there are drawbacks to this method of partitioning. Specifically, there are inevitable plate-to-plate variations in well and chamber volume. More importantly, the filling of wells and chambers is not verified or reported, which means that the dPCR partition volumes may not be accurately known or consistent. Since the accuracy and precision of partition volume relates to the accuracy and precision of dPCR results through Poisson statistics, these microwell and chamber partitioning methods can produce inaccurate and imprecise dPCR results.
