Restriction fragments from different sources can base-pair to form a recombinant DNA.
Many restriction enzymes make staggered cuts and create fragments that have sticky ends.
If fragments that have been cut by the same restriction enzyme are mixed together, the sticky ends will base-pair.
Nicks remain after the molecules base-pair.
The enzyme DNA ligase can seal these nicks to complete the recombinant DNA molecule.
The same restriction enzyme is used to cut both chromosomal DNA and plasmid DNA into fragments.
The DNA fragments are mixed together and modification enzymes are added.
The result is a collection of recombinant plasmids.
The recombinant plasmids are inserted into host cells. These cells can divide rapidly and make additional copies of the foreign DNA.
