Lina Streich is a Predoctoral Fellow, Prevedel Group at EMBL Heidelberg. The Prevedel group develops new optical techniques for investigating dynamic cellular processes deep inside tissue in vivo.

Abstract: The main hurdles for imaging deep into the intact mammalian brain are tissue scattering and the degradation of image quality due to optical aberrations and motion artefacts. In my talk I will present our work on a minimally invasive intravital imaging methodology that adresses these shortcomings by combining three-photon excitation, indirect adaptive optics correction, and active electrocardiogram gating. I will show how our method can be used to achieve near-diffraction limited resolution up to a depth of 1.2mm in the mouse brain to visualize fine structures such as dendrites and spines in deep cortical and subcortical brain regions. Furthermore, I will also demonstrate the capability of our microscope to record calcium transients in deep-layer astrocytes in-vivo.
