Adolescence is a key period of neurodevelopment when children's white matter structure continue to mature, especially frontal lobe connections and association tracts. It is also the period during which psychiatric disorders commonly emerge, and many of them are correlated with white matter abnormalities. Both brain development and psychiatric disorders are known to be significantly heritable. In addition, some genetic effects may only be expressed during particular periods of development.Thus, we hypothesized that genetic risk for psychiatric disorders may alter adolescent white matter development. To test this hypothesis, we analyzed the effect of polygenic risk scores on longitudinal changes in white matter structure for for five major psychiatric disorders: attention deficit hyperactive disorder (ADHD), bipolar disorder (BIP), anxiety disorders (ANX), major depressive disorder (MDD), and schizophrenia (SCZ).We found that polygenic risk scores were strongly correlated with corresponding symptom scales for all disorders except BIP and SCZ, but genetic risk was correlated with change in symptoms for only ANX and MDD.We were unable to find any significant correlations between polygenic risk scores and white matter structure metrics. However, in post-hoc analyses, we found that polygenic risk score for ANX positively moderated the association between mean diffusivity of multiple white matter tracts and the symptom scale.In conclusion, further investigation and additional longitudinal data are necessary to discern the neurobiological mechanisms behind the effect of polygenic risk on change in ANX and MDD symptoms.
